چکیده

Caenorhabditis elegans (C. elegans) as a well-established multicellular model organism has been widely used in the biological field for half century. Its numerous advantages including small body size, rapid life cycle, high-reproductive rate, well-defined anatomy, and conserved genome, made C. one of most successful organisms. Discoveries obtained from have great contributions to research fields such development, aging, biophysics, immunology, neuroscience. Because its transparent giant cell is also an ideal subject high resolution high-throughput optical imaging analysis. During past decade, advances develop biomolecule-targeting techniques noninvasive imaging. These novel technologies expanded toolbox qualitative quantitative analysis biomolecules elegans. In this review, we summarize recently developed fluorescent probes or labeling visualizing at cellular, subcellular molecular scale by using major designed specifically applications Combining technological with will provide new horizons high-efficiency live

Red emission fluorescent probes for visualization of monoamine oxidase in living cells

Here we report two novel red emission fluorescent probes for the highly sensitive and selective detection of monoamine oxidase (MAO) with large Stokes shift (227 nm). Both of the probes possess solid state fluorescence and can accomplish the identification of MAO on test papers. The probe MAO-Red-1 exhibited a detection limit down to 1.2 μg mL(-1) towards MAO-B. Moreover, the cleavage product w...

Cell-trappable fluorescent probes for nitric oxide visualization in living cells.

Two new cell-trappable fluorescent probes for nitric oxide (NO) are reported based on either incorporation of hydrolyzable esters or conjugation to aminodextran polymers. Both probes are highly selective for NO over other reactive oxygen and nitrogen species (RONS). The efficacy of these probes for the fluorescence imaging of nitric oxide produced endogenously in Raw 264.7 cells is demonstrated.

[Development of near-infrared fluorescent probes for in-vivo imaging].

The number of reports on new techniques in molecular imaging has been recently increasing because of their usefulness in biological, medical, and clinical research. Fluorescence imaging methods are generally superior in terms of sensitivity, selectivity and ease of use. Cyanine dyes have been employed as fluorescent labels in fluorescence imaging studies of biological mechanisms. In particular,...

Glycoproteomic probes for fluorescent imaging of fucosylated glycans in vivo.

Glycomics is emerging as a new field for the biology of complex glycoproteins and glycoconjugates. The lack of versatile glycan-labeling methods has presented a major obstacle to visualizing at the cellular level and studying glycoconjugates. To address this issue, we developed a fluorescent labeling technique based on the Cu(I)-catalyzed [3 + 2] cycloaddition, or click chemistry, which allows ...

Fluorescent probes that stain living nerve terminals.

We have evaluated the efficacy of 18 cationic mitochondrial dyes that, as a class, show some ability to stain living nerve terminals. Several of these agents provide excellent staining of neuromuscular junctions in a wide range of species. More detailed studies of the most effective of these dyes--4-(4-diethylaminostyryl)-N-methylpyridinium iodide (4-Di-2-ASP)--indicate that it has no lasting e...
